PlasmoGEM artificial chromosome resource

We have constructed a new linear low-copy P. berghei artificial chromosome (PbAC) vector, which is capable of stably maintaining large fragments of P. berghei genomic DNA. Using the PbAC vector we have generated an arrayed library of P. berghei artificial chromosomes harbouring large P. berghei genomic DNA inserts that provides an off-the-shelf complementation resource with no additional modifications required prior to transfection. Furthermore, PbAC clones can be modified by recombinase-mediated engineering (recombineering, Pfander et al., 2011), e.g. to generate epitope tagging vectors for localisation and expression studies using an exogenous copy of the target genes.
